摘要: Purpose: To evaluate the short-term adverse effects of administration dolastatin-10 (Dol-10) to dogs with spontaneously occurring malignant tumors. Methods: A total 34 tumor-bearing were given Dol-10 as a rapid intravenous bolus every 14 days at starting dosages ranging from 200 350 µg/m2. Acute and effects, antitumor response, duration response characterized. Results: The maximum tolerated dose varied greatly patient patient, but reasonable for further studies was established 300 median number treatments per dog 2 (range 1 17). Granulocytopenia dose-limiting toxicity. overall rate 3%, consisting complete durable (30 months) in high-grade lymphoma that refractory standard therapy. Two minor or transient responses observed, two experienced disease stabilization 8 16 weeks. Conclusions: appears be well doses approaching those by humans. clinical activity observed non-Hodgkin's warrants investigation.
