摘要: Summary A novel bop gene was described from Halobiforma lacisalsi strain AJ5T, an extremely halophilic archaeon isolated Ayakekum Lake, China. Following six rounds of PCR amplification based on the conserved fragment gene, complete sequence including 5′ and 3′ flanking regions fragment, obtained by ligation-mediated (LPA) approach. The data presented provide us with further insight into distribution bop-like genes in family Halobacteriaceae. This is first example a halophiles found high-pH environment. Alignment hydropathy analysis deduced amino acid identified functional sites as well some variations compared other bacterio-opsins. Molecular phylogenetic revealed position bacterio-opsin AJ5, which closest to that Haloterrigena sp. arg-4 85% identity. In presence all-trans retinal, recombinant Escherichia coli cells expressing turned dark purple. purple membrane E. showed maximal absorption at 540 nm.
