摘要: Human ovarian tumors metastasize by direct extension into the peritoneal cavity leading to tumor cell implantation onto surfaces. Successful formation of implants is dependent on ability ascitic cells infiltrate mesothelium, and become firmly adherent underlying extracellular matrix (ECM). In order investigate this process in more detail, an vitro model system was developed employing human mesothelial grown ECM-coated culture dishes. The carcinoma derived from fluid attach monolayer ECM, ECM alone or plastic quantitated with use 51Cr radio-labelled cells. Tumor exhibited a rapid firmer attachment than plastic. Using agitation stimulate dynamics shear forces vivo, arrest found be limited but it occurred at slower rate did without agitation. also restricted areas exposed wounded mesothelium as assessed phase-contrast microscopy. Morphologic alterations induced were observed scanning electron microscopy (SEM) immunohistochemical staining which included disruption intercellular junctions retraction cells, exposure subsequent proliferation ECM. This would appear useful for elucidating mechanisms adhesion proliferation, assessing various therapeutic modalities their block implantation, invasion growth
