摘要: Staphylococcus aureus is an opportunistic pathogen affecting both human and animal species. An effective vaccine to prevent S. bovine disease transmission would have positive effects on well-being, food production, health. The objective of this study was identify multiple antigens that are immunoreactive during udder colonization for exploration as mastitis. produces several cell wall-anchored surface-associated virulence factors play key roles in the pathogenesis Many these proteins conserved between different strains represent promising candidates. We used immunoproteomics approach antigenic from surface aureus. expression wall induced under low iron conditions, followed by trypsin extraction separation 2-dimensional electrophoresis. separated were blotted with antibodies mastitic milk identified liquid chromatography-mass spectrometry. Thirty-eight unique identified, which 8 predicted be exposed involved virulence. Two proteins, iron-regulated determinant protein C (IsdC) ESAT-6 secretion system extracellular (EsxA), cloned, expressed, purified Escherichia coli confirmation immune reactivity ELISA. A PCR 37 isolates indicated presence esxA isdC conserved, amino acid alignments revealed IsdC EsxA sequences highly conserved. technique generated reproducible results reactive further characterization.
