摘要: Grifolin, a secondary metabolite isolated from the fresh fruiting bodies of mushroom Albatrellus confluens, has been shown to induce Cl phase cell-cycle arrest in tumor cells previous studies our group. However, mechanisms action are not completely understood. Our group further demonstrated that grifolin upregulates death-associated protein kinase 1 (DAPK1) nasopharyngeal carcinoma (NPCs). Here, we found induced dephosphorylation DAPK1 (Ser308) activate and subsequent phosphorylation its potential downstream effector p21 (Thr145) cell CNE1. Inhibition by introducing siRNA targeting reversed grifolin- p21. Furthermore, confirmed increased half-life promoted stability. Flow cytometry analysis was involved grifolin-induced CNE1 cells. The similar effects mechanism beneath were identified another HONE1. In addition, observed protein-protein interaction ERK1/2 prevent nucleolus translocation. findings indicate plays crucial role induction at grifolin.
