An in vitro whole blood model to test the effects of different stimuli conditions on the release of myeloperoxidase and elastase by equine neutrophils

摘要: Horses are particularly sensitive and exposed to excessive inflammatory responses evolving toward an important stimulation of polymorphonuclear neutrophils (PMNs). The aim this work was stimulate equine in whole blood evaluate their response by measuring the release total active myeloperoxidase (MPO) elastase, considered as markers neutrophil degranulation. Because critical importance concomitant presence LPS TNF-α pathological situations, we combined these two natural mediators PMN compared with those obtained after each mediator used alone well-known artificial systems such 12-phorbol 13-myristate acetate (PMA) combination cytochalasin B (CB) N-formyl-methionyl-leucyl-phenylalanine (fMLP). All activation systems, PMA, CB/fMLP, TNF-α, LPS/TNF-α, induced a significant MPO but only combinations CB/fMLP LPS/TNF-α significantly favored MPO. Regarding did not observe all stimulated conditions except PMA. It appears clearly that choice model is fundamental for selection potentially pharmacological agents, especially on activity.
