摘要: The von Hippel-Lindau (VHL) disease gene is a novel multiple tumor suppressor which plays causal role in the origin of some common cancers including clear cell renal carcinomas and hemangioblastomas central nervous system. Here we report identification transcription start sites promoter human VHL gene. sequence does not contain TATA CCAAT boxes. Transcription initiated around putative SP1 binding site about 60 bp upstream from first AUG codon mRNA. Several factor sites, notably for nuclear respiratory 1 PAX, were found sites. Promoter-luciferase expression constructs demonstrate, that functional when transfected into 293 cells (transformed primary embryonal kidney cells) UMRC 6 carcinoma cells. Activity dependent on correct orientation promoter. A minimal region 106 was delineated. set minigenes, containing 5' flanking genomic region, constructed In these level minigenes driven by comparable with endogenous expression.
