Effect of erythropoietin, 5-fluorouracil and SN-38 on the growth of DLD-1 cells

摘要: Abstract Supplementation of recombinant human erythropoietin (rHuEpo) is one the methods for treatment anemia. The influence rHuEpo on proliferation or clonogenic growth cancer cells not clear and some published results are conflicting. aim this work was to study effect colon when given alone in combination with cytostatics. Human adenocarcinoma (DLD-1) were cultured medium rHuEpo, 5-fluorouracil (5-FU) an active metabolite irinotecan (SN-38). Cell viability determined using a hematocytometer 0.4% (w/v) trypan blue dye. measured by MTT assay. Expression EpoR, Bax, Bcl-2 Akt1 protein assessed Western blot. indicate dose-dependent inhibitory DLD-1 cell proliferation. Moreover, combined cytotoxic agents such as 5-FU SN-38 increases antitumor action, which indicated decreases test, cell numbers DNA synthesis. We found significant increase expression all grown containing 3 IU rHuEpo. observed that EpoR constitutively expressed DLD-1 cells. Our acts via directly inhibit indirectly modulate cytostatics effects SN-38.
