摘要: We determined the effects of follicular fluid in maturation medium on bovine oocyte maturation, fertilization and subsequent development, as well number cells blastocysts following culture. Fluid oocytes from follicles less than 5 mm diameter were collected ovaries slaughtered cows. For medium, at concentrations 10, 30 or 60% (v/v) was added to Medium 199 with Earle's salts supplemented 0.1 microg/ml estradiol-17 beta (E(2), Experiment 1) E2 100 IU/ml hCG (Experiment 2). The control contained polyvinylpyrrolidone (PVP; 3 mg/ml) instead fluid. After for 24 h, fertilized vitro bull frozen-thawed spermatozoa cultured a monolayer granulosa 9 d. There no differences rates oocytes. In 1, containing 10% did not affect developmental rate > 2-cell, 8 16-cell, blastocyst hatched stage embryos, respectively; whereas decreased embryonic development (P < 0.05) compared control. Blastocysts developed which had been matured 10 30% fluid/E(2) more controls 0.01). 2, control, but Day E(2) + hCG. results these experiments suggest that addition enhances cell numbers vitro.
