Electrochemical lactate biosensor based upon chitosan/carbon nanotubes modified screen-printed graphite electrodes for the determination of lactate in embryonic cell cultures.

摘要: l-lactate is an essential metabolite present in embryonic cell culture. Changes of this important during the growth human embryo reflect quality and viability embryo. In study, we report a sensitive, stable, easily manufactured electrochemical biosensor for detection lactate within cultures media. Screen-printed disposable electrodes are used as sensing platforms miniaturization biosensor. Chitosan/multi walled carbon nanotubes composite have been employed enzymatic immobilization oxidase enzyme. This novel analytical efficacy explored towards model (buffer) solutions found to exhibit linear response over concentration range 30.4 243.9 µM phosphate buffer solution, with corresponding limit (based on 3-sigma) 22.6 exhibits sensitivity 3417 ± 131 µAM(-1) according reproducibility study. These biosensors high reproducibility, relative standard deviation less than 3.8% 82% after 5 months stored at 4 °C. Furthermore, performance liquid chromatography technique has utilized independently validate determination commercial culture medium providing excellent agreement between two protocols.
