Theoretical Study on the Cyclization Mechanism of Dipeptides

摘要: Cyclization is an important chemical reaction for the dipeptides containing N-alkyl groups. The cyclization mechanism has been examined by theoretical calculations. Our calculation results indicate that most favorable piperidine-catalyzed stepwise mechanism, in which piperidine acts as a proton shuttle. attack of N-terminal amino nitrogen at C-terminal carbonyl carbon along with transfer rate-limiting step. effect alkyl substituent on amide N was then examined. Finally, influence solvation effect, electronic and steric investigated. It found all these effects contribute to cyclization.
