摘要: Background The present study was undertaken to examine the ability of lipopolysaccharide-containing outer membrane vesicles (OMV-LPS) and purified LPS (P-LPS) from same meningococcal strain induce expression Toll-like receptors (TLR2 TLR4) TNF-alpha production in leukocytes, further involvement TLRs, CD14 monocyte TNF- alpha an ex vivo human whole blood system. Material/methods OMV-LPS or P-LPS were added TLR2/4 leukocytes measured by flow cytometry. To TLRs cytokine we used monoclonal antibodies against CD14. Results induced surface (maximal at 120 min) TLR2 TLR4 on granulocytes monocytes. incorporated OMV less potent (weight basis) than inducing production. When OMV-LPS, directed caused 45 78% inhibition, respectively. P-LPS, had no effect, whereas anti-TLR4 63% inhibition. Antibodies inhibited nearly completely response irrespective whether presented form vesicles. Conclusions monocytes granulocytes. Surface are essential for vitro cellular activation but functional significance these during infections remains elusive.
