Differential gene expression before and after ionizing radiation of subcutaneous fibroblasts identifies breast cancer patients resistant to radiation-induced fibrosis

摘要: Abstract Background and purpose Differentially gene expression between patients with either very low or high risk of radiation-induced fibrosis (RIF) in patient-derived fibroblasts after irradiation has previously been reported. In the present study, we are investigating robustness changes fibroblasts, whether differential is more pronounced when looking at fold induction levels, taking into account differences background levels patients, there a linear correlation individual RIF fibroblasts. Material methods Gene was analysed by quantitative real-time PCR before fractionated scheme 3×3.5Gy/3 days derived from 26 breast cancer treated post-mastectomy radiotherapy. Results Robust were observed, being most for level (‘after' value divided ‘before' each patient). When including intermediate risk, no genes investigated. Rather, could divide two clearly separated groups, larger, sensitive group smaller resistant group. Conclusions Differential irradiated might be an important tool identification genetic variable RIF, new targets prevention intervention fibrotic process.
