摘要: The pathogenic yeast, Candida albicans, is insensitive to the anti-mitotic drug, benomyl, and dihydrofolate reductase inhibitor, methotrexate. Genes responsible for intrinsic drug resistance were sought by transforming Saccharomyces cerevisiae, a yeast sensitive both drugs, with genomic C. albicans libraries screening on benomyl or Restriction analysis of plasmids isolated from benomyl- methotrexate-resistant colonies indicated that phenotypes encoded same DNA fragment. Sequence showed fragments nearly identical contained long open reading frame 1694 bp (ORF1) small ORF 446 (ORF2) within ORF1 opposite strand. By site-directed mutagenesis, it was shown phenotypes. protein had no sequence similarity any known proteins, including β-tubulin, reductase, P-glycoprotein multi-drug family. gene detected in several strains stellatoidea hybridization polymerase chain reaction.
