摘要: Pre-eclampsia (PE) is a worldwide pregnancy-related disorder. It mainly characterized by defect migration and invasion of trophoblast cells. Recently, circular RNAs (circRNAs) have been believed to play vital role in PE. The expression patterns the biological functions circRNAs PE remain elusive. Here, we performed circRNA microarray identify putative PE-related circRNAs. Bioinformatics analyses were used screen which potential relationships with pre-eclampsia, identified novel (circVRK1) that was up-regulated placenta tissues. By using HTR-8/SVneo cells, circVRK1 knockdown significantly enhanced cell abilities, as well epithelial-mesenchymal transition (EMT). Mechanistically, found PTEN could function ceRNAs miR-221-3p. Overexpression miR-221-3p promoted migration, EMT via regulating PTEN. cotransfection inhibitor or reversed effect from knockdown. Moreover, circVRK1/miR-221-3p/PTEN axis greatly regulated Akt phosphorylation. In general, suppresses EMT, acting ceRNA regulate PTEN, further inhibit PI3K/Akt activation. purpose this paper open wide insights investigate onset provide new therapeutic targets
