摘要: Electroporation is well known to depend non-linearly on the magnitude and duration of change ΔU(t) in transmembrane voltage. In case cell suspension experiments, an electric field Ee(t) within electrolyte causes ΔU(t), which governed by both size shape a cell, also Ee(t). It therefore important determine time dependence cells are actually exposed electroporation experiments. This can be significantly different from nominal En, calculated using electrode voltages geometries alone. Throughout we used single, nominally exponential pulses with constants τpulse ranging about 0.6 5 ms found that Ee was always less than En. order actual pulse, measured voltage across electrodes, current through cuvette, temperature rise pulsing medium, two special electrodes placed cuvette. From these measurements strength inside cuvette methods. addition, compared expected electrical power dissipation. some cases there much larger (“anomalous”) heating, due interfacial electrochemical heat production; for one solution Te(t) 30 K expected. These effects experiments aimed at elucidating mechanism, comparing results obtained under conditions, guiding applications.
