Preparation of kenaf stem hemicellulosic hydrolysate and its fermentability in microbial production of xylitol by Escherichia coli BL21

摘要: Kenaf (Hibiscus cannabinus L.), a potential fibre crop with desirably high growth rate, could serve as sustainable feedstock in the production of xylitol. In this work, extraction soluble products kenaf through dilute nitric-acid hydrolysis was elucidated respect to three parameters, namely temperature, residence time, and acid concentration. The study will assist evaluating performance terms xylose recovery. result point out that maximum yield 30.7 g per 100 g dry attained from 2% (v/v) HNO3 at 130 °C for 60 min. detoxified hydrolysate incorporated primary carbon source subsequent fermentation by recombinant Escherichia coli strain on five different semi-synthetic media xylitol were evaluated herein. Among these media, batch cultivation basal salt medium (BSM) afforded highest 0.35 g/g based consumption, which corresponded 92.8% substrate utilization after 38 h. Subsequently, E. xylose-based supplemented BSM resulting 6.8 g/L corresponding 0.38 g/g. These findings suggested use be advantageous development production.
