摘要: The uptake of adenosine by an kinase deficient variant C1300 murine neuroblastoma cells has been studied in the absence and presence erythro-9-(2-hydroxy-3-nonyl)adenine, a potent adenine deaminase inhibitor. Although 100 μM inhibitor completely blocks metabolism under conditions studied, is concentrative, i.e., intracellular concentration exceeds extracellular concentration. This concentrative effect decreases as increases hypothesized to be due binding component. Despite this effect, we believe that kinetics uptake, determined experiments with short (10–20 s) periods, reflect transport facilitated diffusion process. nucleoside system appears nonspecific competitively antagonized thymidine. It does not appear necessary inhibit order study these Km for affected erythro-9-(2-hydroxy-3-nonyl)adenine. However, erythro-9-(2-hydroxy-3-nonyl)adenine depress V transport. probably inhibition thymidine similarly affected.
