摘要: TSP1 was reported to be involved in multiple biological processes including the activation of TGF-β signaling pathways and regulation angiogenesis during wound repair tumor growth, while its role ovarian folliculogenesis remains elucidated. In present study, Tsp1a found expressed oogonia granulosa cells phase I IV follicles ovaries Nile tilapia by immunofluorescence. tsp1a homozygous mutants were generated CRISPR/Cas9. Mutation resulted increased oogonia, reduced secondary growth delayed ovary development. Expression cell proliferation marker PCNA significantly up-regulated mutant ovaries. Furthermore, transcriptomic analysis revealed that expressions DNA replication related genes up-regulated, cAMP MAPK pathway which inhibit promote differentiation down-regulated. addition, aromatase (Cyp19a1a) expression serum 17β-estradiol (E2) concentration decreased mutants. These results indicated lacking inhibited turn, E2. Taken together, our essential for tilapia.
