A single-chain antibody fragment specific for the Plasmodium berghei ookinete protein Pbs21 confers transmission blockade in the mosquito midgut.

摘要: Mouse monoclonal antibody 13.1 (mAb 13.1) directed against Pbs21, a 21-kDa sexual-stage surface protein of Plasmodium berghei, is known to inhibit oocyst development from gametocytes and ookinetes in the mosquito midgut. To examine properties potential uses single-chain fragment (scFv) for blocking transmission malaria parasites mosquitoes, we have cloned sequenced genes encoding variable regions immunoglobulin heavy light chains (VH VL) mAb 13.1. The VH VL were assembled as an scFv gene, expressed baculovirus expression system. Following purification scFv, Western blotting inhibition ELISA assays confirmed that retained binding specificity parent Pbs21. Furthermore, bound P. berghei ookinetes, blocked midgut by at least 93%, assessed counts mosquitoes. We suggest gene could be useful not only studying mechanism blockade, but also generating, germline transformation, model system evaluate production mosquitoes refractory malaria.
