摘要: Purpose To investigate the change of IL-10 producing regulatory B cells (Bregs), which function to suppress peripheral immune responses, in patients with thyroid associated ophthalmopathy (TAO). Methods Peripheral blood mononuclear (PBMCs) were isolated from healthy controls (N=54), Grave's disease (N=26), and TAO (N=125), stimulated CpG/CD40L. The frequency Bregs expression response thyroid-stimulating hormone (TSH) stimulation measured by flow cytometry. CD4+ T cultured Breg-depleted PBMCs elucidate patients. potential immunoregulatory mechanism was also investigated western blot chromatin immunoprecipitation assays. Results Active had higher baseline levels their than both inactive TSH promoted Bregs. defective suppressing activation IFN-γ+ IL-17+ vitro. Conclusions We found that are functionally defective, suggesting might be responsible for pathogenesis TAO.
