The level of mRNA encoding the amphotropic retrovirus receptor in mouse and human hematopoietic stem cells is low and correlates with the efficiency of retrovirus transduction

摘要: Abstract The low level of amphotropic retrovirus-mediated gene transfer into human hematopoietic stem cells (HSC) has been a major impediment to therapy for diseases. In the present study, we have examined retrovirus receptor (amphoR) and ecotropic mRNA expression in highly purified populations mouse HSC. Murine HSC with undetectable levels amphoR relatively high were studied. When these analyzed simultaneously transduction, provirus sequences detected 10 13 long-term repopulated animals, while proviral only one recipient. A second distinct population murine isolated that express 3-fold higher mRNA. 11 mice contained 6 sequences, significant increase transduction (P = 0.018). These results indicate that, among heterogeneous adult bone marrow, subpopulation highest is more efficiently transduced by retrovirus. related found (CD34+ CD38-) committed progenitor CD38+).
