A new lineage of trypanosomes from Australian vertebrates and terrestrial bloodsucking leeches (Haemadipsidae)

摘要: Little is known about the trypanosomes of indigenous Australian vertebrates and their vectors. We surveyed a range blood-feeding invertebrates for by parasitological PCR-based methods using primers specific to small subunit ribosomal RNA (SSU rRNA) gene genus Trypanosoma. Trypanosome isolates were obtained in culture from two common wombats, one swamp wallaby an bird (Strepera sp.). By PCR, blood samples three brush-tailed wallaby, platypuses frog positive trypanosome DNA. All blood-sucking screened negative both microscopy except specimens terrestrial leeches (Haemadipsidae). Of latter, Micobdella sp. Victoria 18 Philaemon Queensland PCR. Four Haemadipsa zeylanica Sri Lanka Leiobdella jawarerensis Papua New Guinea also PCR sequenced SSU rRNA glycosomal glyceraldehyde phosphate dehydrogenase (gGAPDH) genes order determine phylogenetic positions new vertebrate leech trypanosomes. In trees based on these genes, fell several distinct clades, mostpart being more closely related outside Australia than each other. Two previously undescribed clade with Trypanosoma theileri, cosmopolitan bovid trypanosome, cyclops Malaysian primate. The trypanosomes, T. frog. suggest that haemadipsid may be significant widespread vectors Asia.
