Maintenance of Acetylcholine Receptor Number by Neuregulins at the Neuromuscular Junction in Vivo

摘要: ARIA (for acetylcholine receptor-inducing activity), a protein purified on the basis of its ability to stimulate receptor (AChR) synthesis in cultured myotubes, is member neuregulin family and present at motor endplates. This suggests an important role for neuregulins mediating nerve-dependent accumulation AChRs postsynaptic membrane. Nerve-muscle synapses have now been analyzed neuregulin-deficient animals. Mice that are heterozygous deletion isoforms containing immunoglobulin-like domain myasthenic. Postsynaptic AChR density significantly reduced, as judged by decrease mean amplitude spontaneous miniature endplate potentials bungarotoxin binding. On other hand, evoked was not decreased, due increase number quanta released per impulse, compensation has observed myasthenic states. Thus, membrane depends immunoglobulin-containing throughout life animal.
