摘要: Cardiovascular diseases (CVD) represent one of the leading causes mortality worldwide. Epidemiological studies have suggested an inverse association between consumption citrus fruit and risk CVD. However, molecular mechanisms by which flavonoids contribute to cardiovascular health not been fully elucidated. In addition, evidence from human is limited contradictory. Therefore, aims this thesis were investigate impact on endothelial cells their effects function in acute manner following regular consumption. The potential selected modulate gene expression relevant nitric oxide (NO) production antioxidant status was investigated EA.hy926 cells; effect flavanone-rich blond orange juice (total flavanone content=81.7 mg/100 ml) vascular (cardiac electrical activity, blood pressure, compliance, peripheral flow, pulse transit time wave velocity) healthy subjects (n=13) determined; a randomized controlled crossover trial, content=89.7 function, lipid profile, inflammation overweight/obese men premenopausal women European origin (n=15) examined 2-week (400 ml/d) sugar-matched control drink. The vitro study demonstrates NOS3 mRNA protein significantly upregulated nobiletin tangeretin; incubation with also resulted significant reduction ET-1 levels induction HO-1 GCLC. Vascular related markers acutely altered at 2 h post under conditions. Following juice, improved 8.15% ± 2.92% 10.16% 3.31%, compared drink (time treatment interaction: P=0.002). Interestingly, favourable only observed overweight but obese subjects. Baseline hsCRP generally within ranges during supplementation. Present data indicate that exerts beneficial origin, may be attributed elevated NO bioavailability cell-protective enzymes as evidenced findings.
