Identification and Quantitation of Unsaturated Fatty Acid Isomers by Electrospray Ionization Tandem Mass Spectrometry: A Shotgun Lipidomics Approach

摘要: Identification and quantification of unsaturated fatty acid (FA) isomers in a biological system are significant the study lipid metabolism catabolism, membrane biophysics, pathogenesis diseases but challenging lipidomics. We developed novel approach for identification quantitation FA by exploiting two facts: (1) anions yield fragment ion(s) from loss CO(2) or H(2)O upon collision-induced dissociation; (2) ions yielded discrete have distinct profiles ion intensity vs. collision conditions. These likely result differential interactions negative charge with electron clouds double bonds due to their different distances isomers. The was also extended analyze bond chains present phospholipids multistage tandem mass spectrometry. Collectively, we new endogenous species intact phospholipid species. believe that this should further advance lipidomic power biochemical mechanisms underlying metabolic diseases.
