Protective influenza-specific CD8 T cell responses require interactions with dendritic cells in the lungs

摘要: Influenza infections induce a rapid, but transient, dendritic cell (DC) migration from the lungs to lymph nodes (LNs) that is followed by substantial recruitment of DCs into without subsequent LNs. Given peripheral are primarily thought be involved in initiation adaptive immunity after lymphoid tissues, what role these newly lung-recruited play influenza virus unclear. In this study, we demonstrate loss non-LN migratory pulmonary DC subsets increases mortality, sustains higher viral titers, and impairs CD8 T responses. Reconstitution with plasmacytoid DCs, CD8α+ or interstitial restores responses contact–, major histocompatability complex I–, peptide–dependent manner. Thus, their initial activation LN, protective influenza-specific require additional antigen-dependent interactions, specifically lungs.
