摘要: Collagen is the most abundant extracellular matrix (ECM) protein in human body, thus widely used tissue engineering and subsequent clinical applications. This study aimed to extract collagen from ovine (Ovis aries) Achilles tendon (OTC), evaluate its physicochemical properties potential fabricate thin film with fibrils a random or aligned orientation. Acid-solubilized was extracted using 0.35M acetic acid, 80% of measured as collagen. SDS-PAGE mass spectrometry analysis revealed presence alpha 1 2 chain type I (col I). Further Fourier transform infrared (FTIR), X-ray diffraction (XRD) energy dispersive spectroscopy (EDS) confirms triple helix structure col I, similar commercially available rat tail I. Drying OTC solution at 37°C resulted formation randomly orientated (random film; RCF). Introduction unidirectional mechanical intervention platform rocker prior drying facilitated fabrication orientation fibril (aligned ACF). It shown that both RCF ACF significantly enhanced dermal fibroblast (HDF) attachment proliferation than on plastic surface. Moreover, cells were distributed RCF, but direction ACF.
