摘要: The re-emergence of Zika virus (ZIKV) caused widespread infections that were linked to Guillain-Barre syndrome in adults and congenital malformation fetuses, epidemiological data suggest ZIKV infection can induce protective antibody responses. A more detailed understanding anti-ZIKV responses may lead enhanced discovery improved vaccine designs against related flaviviruses. Here, we applied recently-invented library-scale screening technologies determine comprehensive functional molecular genetic profiles naturally elicited human antibodies three convalescent individuals. We leveraged natively paired yeast display NGS predict cross-reactivities coarse-grain affinities, perform in-depth immune profiling IgM, IgG, IgA repertoires peripheral blood, reveal maturation state-dependent interactions. Repertoire-scale comparison VLP-specific non-specific the same individuals also showed mean somatic hypermutation levels substantially influenced by donor-intrinsic characteristics. These provide insights into antiviral disease outline systems-level strategies track emergent viral infections.
