摘要: Endothelial cells (ECs) isolated from endothelial progenitor in blood have great potential as a therapeutic tool to promote vasculogenesis and angiogenesis treat cardiovascular diseases. However, current methods isolate ECs are limited by low yield with few colonies appearing during isolation. In order utilize blood-derived for applications, simple method is needed that can produce high of small volumes without the addition animal-derived products. For first time, we show human be prior separation components through technique diluted whole incubation (DWBI) utilizing commercially available serum. We (~10 mL) via DWBI characterized them flow cytometry, immunohistochemistry, uptake DiI-labeled acetylated density lipoprotein (DiI-Ac-LDL). These functional demonstrated their ability form tubular networks Matrigel, adhere align under physiological fluid shear stress, increased nitric oxide flow. An average 7.0 ± 2.5 EC passed all tests described above were obtained per 10 mL compared only 0.3 ± 0.1 traditional based on centrifugation. The time until colony appearance was 8.3 ± 1.2 days 12 ± 1.4 days isolation method. A simplified method, such DWBI, combination advances could enable use clinical practice.
