VİRAL HEPATİTLERDE MATRİKS METALLOPROTEİNAZLARIN VE İNHİBİTÖRLERİNİN EKSPRESYON PATERNLERİ VE BUNLARIN FİBROZİS İLE İLİŞKİLERİ

摘要: Amac: Bu arastirmanin amaci, kronik viral hepatitli hastalarin karaciger biyop- silerinde MMP ve TIMP ekspresyonlarinin siddeti yayginligi ile fibrozis ev- resinin nekroinflamatuar derecesinin iliksisini saptamaktir. Gerec Yontem: 67 hastanin biyopsilerinde MMP’lerin ekspresyon yayginligini, siddetini lokalizasyonunu degerlendi- rip, TIMP-1 TIMP-2 korelasyonunu arastirdik. Yirmiyedi B he- patitli, 40 C biyopsileri 5 normal kont- rol biyopsisi immunhistokimyasal olarak MMP-2, MMP-9, boyandi. Hastalara ait klinik bilgiler dosyalarindan elde edildi. Bulgular: HBV veya HCV iliskisinden bagimsiz MMP-9 ekspresyo- nu hepatositlerde cok az oranda (%3) iken, MMP-2 olgularin %43’ unde hepa- tositlerde portal alanlardaki igsi hucrelerde eksprese oldu. TIMP- 2 hepatositlerde, sinuzoidal degisen oranlarda (sirasiyla %12 21) boyanma goruldu. Sadece eks- presyonu ekspresyonu koreleydi (p=0,01). Diger yandan ne de ekspresyonlari inflamasyonun fibrozi- sin korelasyon gostermedi. Sonuc: TIMP’ lerin kan seviyeleri siroz gelisiminin korele oldugu bircok grup tarafindan gosterilmis olsa da bizim bulgularimiza gore patit orneklerindeki patern- lerindeki degisiklikler serumdaki konsantrasyon degisikliklerini aciklamaz hastaligin ilerledigini gostermez. nedenle kandaki seviylerinin karacigere spesifik net degildir. EXPRESSION PATTERNS OF MATRIX METALLOPROTEINASES AND THEIR INHIBITORS IN VIRAL HEPATITIS RELA- TION WITH FIBROSIS ABSTRACT Purpose: To determine the association between expression patterns of mat- rix metalloproteinases (MMPs) and extent fibrosis inflammation in chro- nic hepatitis. Materials Methods: We examined localization correlated data with their main inhibitors, tissue inhibitors (TIMP-1 TIMP-2), chronic patitis patients. Immunohistochemistry was performed for MMPs TIMPs liver biopsies controls, 27 patients hepatitis B, C. Results: In hepatitis, hepatocytes very small (3%), whereas expressed some spindle cells tracts 43% cases. Variable degrees staining were ob- served hepatocytes, sinusoidal cells, tract (12% 21%, respectively). Only (p=0.01). On other hand, neither nor expressions showed any correla- tion fibrosis.Conclusion: Our findings sho- wed that alterations TIMP-1, samples do not explain changes se- rum concentrations mirror progression disease. Thus, it is still unclear whether circulating levels are specific. Key Words: Chronic Hepatitis, Fibrosis, Staging, MMPs, TIMPs.
