摘要: Neuroglobin (Ngb), a recently discovered globin, is predominantly expressed in the brain, retina, and other nerve tissues of vertebrates. The unfolding processes apo-neuroglobin (apoNgb) induced by guanidine hydrochloride (GdnHCl) urea were investigated spectroscopic methods. In processes, apoNgb's tertiary structural transition was monitored changes intrinsic fluorescence emission spectra, its secondary measured far-ultraviolet circular dichroism (CD) spectra. addition, 8-anilino-1-naphthalenesulfonic acid (ANS), hydrophobic cluster binding dye, also used to monitor process apoNgb explore intermediates. Results showed that GdnHCl-induced via three-state pathway, is, Native state → Intermediate Unfolded , during which intermediate inferred an increase intensity change CD value. Gibbs free energy are 10.2 kJ·mol−1 for first 14.0 second transition. However, urea-induced only underwent two-state transition: Partially unfolded . result GdnHCl can efficiently affect conformational states compared with those urea. work will benefit have understanding mechanism
