摘要: This investigation was undertaken to explain the possible, mechanism(s) of protection by diethyldithiocarbamate (DDTC) against cisplatin ototoxicity. Male Wistar rats (250-275 g) underwent pretreatment auditory brain stem-evoked responses (ABRs). The different groups were injected as follows: (1) (16 mg/kg i.p.), (2) plus DDTC i.p. + 600 mg/kg, s.c.), and (3) control rats. Post-treatment ABRs performed after 3 days euthanized cochleae harvested. analyzed for glutathione (GSH) oxidized glutathione, HPLC, activities antioxidant enzymes, malondialdehyde levels, spectrophotometry. cisplatin-injected showed a threshold elevation 36 +/- 3.05 dB above thresholds using click stimulus. Rats treated with then did not show significant hearing threshold. DDTC-mediated associated higher levels GSH (0.81 0.11 nmol/mg tissue), compared 0.45 0.02 tissue following administration alone. Administration restored cochlear GSH-Px activity level. Cisplatin-treated found have decreased (75% control).
