摘要: UNLABELLED Leucovorin (LV)/5-fluorouracil combination is one of the first-line forms chemotherapy for colorectal cancer, and in this regimen it was recommended that 5-FU be infused continuously from venous route. However, since limits patients' ADL, oral administration preferable. The purpose study to elucidate mechanisms orally-administered LV/5-FU. MATERIAL AND METHODS Gastrostomy performed, 1 x 10(6) colon 26 transplanted subcutaneously CDF1 mice. In continuous group, either 10, 20, 40 mg/kg 100 LV were 7 days following gastrostomy using a microinfusion pump (n = 6). single infusion once day after concomitant with other 6 animals served as control, alone group. Tumor volume, thymidylate synthase inhibition rate (TSIR), incorporation into RNA (F-RNA) body weight measured at end treatment. During experimental period, mice given free access chow water. RESULTS tumor volume suppression significantly higher along amount 5-FU. there no significant difference between groups. TSIR group dose 20 mg/kg. contrast, F-RNA Significant loss not recognized any CONCLUSION From these data, more effective dysfunction. On hand, damaging DNA duplication. summary, mechanism antitumor effect differs method, important understand mechanism.
