摘要: Abstract Lipidomics focuses on the comprehensive analysis of lipids and their interactions with other molecules. Many biological samples are available in small volumes or limited amounts display very complex lipid compositions; thus, highly sensitive lipidomic methods often needed. NanoLC-MS offers extremely high sensitivity, although it is known to be technically more challenging than conventional LC-MS approach, requiring greater care maintenance. This work describes development optimization a nanoLC-MS method for routine lipidome samples. We focused achieving robust conditions sensitivity The nanoLC method, mass spectrometry sample preparation by liquid-liquid extraction were fully optimized using serum deuterated standards, including an evaluation contamination sources. performance was assessed through human pig sera, as well cerebrospinal fluid from pigs. allowed detection 9900 12,200 features employing only 1.0–2.5 μL identification 5842 36 subclasses within 50-min gradient. can easily adapted types where available.
