摘要: Current techniques used to measure lengths of DNA fragments in atomic force microscope (AFM) images require a user operate interactive software and execute tedious error-prone cursor selections. An algorithm is proposed which provides an automated method for determining fragment from AFM without interaction the computer operator (e.g. selections or mouse clicks). The approach utilises image processing tailored characteristics fragments. measurements have mean absolute deviation less than 1 pixel when compared manual image-based measurements. length determined histogram calculated accurate within 3% actual solution. For that are 250 base-pairs long, precision estimated be 17 nm, about 20% total length. This was confirmed easily resolved one differed by only nm. Fragment sizes up 2000 been tested successfully sized. being developed as part new solid-state sizing technique applications such genotyping construction physical genome maps.
