摘要: Methods are described for growing Spirillum lipoferum in quantities sufficient to serve as inoculant field trials of its associative N(2)-fixing ability with higher plants and a source cells the preparation nitrogenase, cytochromes, respiratory enzymes, etc. A heavy inoculum S. grown on NH(4) was transferred medium minimal nitrogen content, initial rapid growth at expense residual combined replaced later by slower N(2). Conversion N(2) fixation prompt upon exhaustion fixed nitrogen; most pO(2) 0.005 0.007 atm. Numbers can be estimated diluting soil, crushed roots, or other material, inoculating diluted samples into stagnant semisolid medium. Development characteristic subsurface layer organisms demonstration these reduce C(2)H(2) presumptive evidence that they lipoferum. With most-probable-number tables observations converted numbers samples. The method indicated may increase 100-fold more roots maize removed from plant incubated 24 h 30 degrees C initially adjusted 0.01
