摘要: The Norway lobster, Nephrops norvegicus, is a burrowing decapod with rhythmic burrow emergence (24 h) governed by the circadian system. It an important resource for European fisheries and its behavior deeply affects availability. current knowledge of biology phenomenological as it currently case almost all crustaceans. In attempt to elucidate putative molecular mechanisms underlying gene regulation in Nephrops, we used transcriptomics approach on cDNA extracted from eyestalk, structure playing crucial role controlling decapods. We studied 14 male lobsters under 12–12 light-darkness blue light cycle. Hiseq 2000 Illumina platform sequence two eyestalk libraries (under darkness conditions) obtaining about 90 millions 100-bp paired-end reads. Trinity was de novo reconstruction transcriptomes; size at which half assembled bases reside contigs (N50) equal 1796 (light) 2055 (darkness). found list candidate clock genes focused our attention canonical ones: timeless, period, bmal1. cloning fragments validated outputs. showed high levels identity (blastx NCBI) known crustacean homologs such Eurydice pulchra (period: 47%, timeless: 59%, bmal1: 79%) Macrobrachium rosenbergii (clock: 100%). also vertebrate-like cryptochrome 2. RT-qPCR that only timeless had robust diel pattern expression. Our data are accordance clock, reinforcing idea clockwork this group shows some differences established model Drosophila melanogaster.
