摘要: Twelve different human primary and metastatic Ewing's sarcoma (ES) primitive peripheral neuroectodermal tumour (pPNET) cell lines were examined by fluorocytometric analysis for the expression of alpha 1, 2, 3 6 very late antigen (VLA) beta 1-integrins. VLA-alpha was abundantly expressed on all typical ES pPNET lines, while absent from atypical (large cell) cells. 2 displayed some lines. In two derived same patient, considerably higher cells compared with exclusively Expression than Adhesion assays purified extracellular matrix (ECM) proteins, using monospecific adhesion-blocking antibodies, disclosed VLA-1 (alpha 1 1) cells, VLA-2 as dual collagen type IV (COIV)/laminin (LM) binding sites, VLA-6 a specific LM site. Treatment up to 48 h recombinant interferon-gamma (rhIFN gamma) necrosis factor-alpha (rhTNF alpha) upregulated expression, concomitant an increase in adhesion COIV LM. Alternatively, these cytokines downregulated decrease conclusion, findings suggest that difference repertory CO integrin receptors reflects status degree differentiation. Furthermore, our data indicate IFN gamma- TNF alpha-mediated alteration level distinct VLAs is correlated changes adhesive behaviour
