摘要: A lectin-impregnated gel was fabricated at the channel crossing point in a microfluidic chip made from polymethyl methacrylate (PMMA). The acrylamide containing lectin photopolymerized to form round (radius 60 μm) by irradiation with an argon laser, which also used for fluorometric detection. This applied specific concentration, elution, and electrophoretic separation of fluorescent-labeled oligosaccharides. Because polyacrylamide mechanically immobilized, it maintained its activity. trap up few tens femtomoles oligosaccharides labeled 8-aminopyrene-1,3,6-trisulfonic acid 2 min factor >800, amount trapped corresponded ca. 70% gel. were released lowering pH acidic background electrolyte. that eluted as broad band concentrated transient isotachophoresis stacking using sodium borate buffer (pH 11.0). stacked sample components then separated fluorometrically detected end channel. Under optimized conditions, resolution saccharides good, similar obtained pinched injection. method preconcentration analysis derived some glycoproteins.
