Transfection of C6 glioma cells with connexin 43 cDNA: analysis of expression, intercellular coupling, and cell proliferation.

摘要: C6 glioma cells express low levels of the gap junction protein connexin 43 and its mRNA display very weak dye coupling. When implanted into rat cerebrum, these quickly give rise to a large glioma. To investigate role junctions in tumor characteristics cells, we have used Lipofectin-mediated transfection introduce full-length cDNA encoding 43. Several transfected clones were obtained that exhibited various amounts transcribed from inserted cDNA. Immunocytochemical analysis revealed an increase amount immunoreactivity being localized at areas intercellular contact as well cytoplasm. The level coupling was also assessed found correlate with mRNA. cell proliferation followed over several days, expressing grew more slowly than non-transfected cells. These will be useful examining tumorigenesis.
