摘要: Possible mechanisms of cytochrome P450 self-inactivation during catalytic turnover have been considered. Two ways hemoprotein inactivation are so far known. The firts, studied extensively by many authors, is the formation active substrate intermediates, capable modifying heme and apoenzyme. second way, revealed quite recently resulting from uncoupled P450-catalyzed monooxygenase reactions, yet to be clarified. Briefly, it involves hydrogen peroxide in center, which interacts with enzyme associated Fe2+, thereby generating hydroxyl radicals that bleach modify This mechanism operates substrates forms partially coupled thus causing as a byproduct.
