Trinitrobenzenesulphonic acid colitis alters Nav 1.8 channel expression in mouse dorsal root ganglia neurons

摘要: Visceral inflammation evokes hyperexcitability in nociceptive dorsal root ganglia (DRG) neurons and these changes are associated with increased voltage-gated sodium channel (Na(v)) 1.8 current density, but the molecular determinants of unclear. This study used Western blotting to measure Na(v) 1.7, 1.9 protein expression during trinitrobenzenesulphonic acid (TNBS) colitis quantitative polymerase chain reaction (PCR) examine corresponding mRNA. Colonic were labelled retrograde tracer Fast Blue injected into wall distal colon PCR performed on laser-captured colonic from at T9-13 or unlabelled DRG upper spinal cord. Immunohistochemistry western blots whole same sites. Blue-labelled demonstrated immunoreactivity. On day 7 colitis, which correlated electrophysiological studies, there was a threefold increase T9 13, 1.7 levels unchanged. There no change alpha-subunit mRNA levels. However, days 2 4, decreased 10-fold. unchanged isolated cord, where not found. These findings suggest that TNBS evoked currents is numbers channels. The absence transcript suggests translational post-translational mechanism, 10-fold recovery preceding this time point also demonstrates complex transcriptional regulation.
