摘要: The nucleotide sequence of the gene coding for xylose isomerase from Ampullariella sp. strain 3876, a gram-positive bacterium, has been determined. A clone fragment 3876 DNA activity was identified by its ability to complement isomerase-defective Escherichia coli strain. One such complementation positive fragment, 2,922 nucleotides in length, sequenced entirety. There are two open reading frames 1,182 and 1,242 on opposite strands this each which could code protein expected size isomerase. 1,182-nucleotide frame as analysis amino-terminal region selected internal peptides. initiates with GTG high guanine cytosine content (70%) an exceptionally strong preference (97%) or third position codons. codes 43,210-dalton polypeptide composed 393 amino acids. is similar other bacterial isomerases limited acid homology available sequences E. coli, Bacillus subtilis, Streptomyces violaceus-ruber. In all cases yet studied, xylulose kinase downstream We present evidence suggesting that these genes similarly arranged.
