摘要: The extent of oxidative DNA damage is considered a biomarker carcinogenic process and could be investigated in population studies using easily obtained cells. oxidized base adduct 8-hydroxy-2-deoxyguanosine (8-OHdG) released by enzymatic hydrolysis commonly assayed high performance liquid chromatography with electrochemical detection. It expressed as ratio 8-OHdG to unoxidized deoxyguanosine. We modified improved this method, determined the optimal time for harvesting buccal mucosa cells (BMC), assessed whether they mirror peripheral circulating blood cell damage, compared anticoagulants, heparin, EDTA consistency measurement leukocyte 8-OHdG. Thirty-one healthy participants, randomized into two groups, donated BMC samples. Samples were collected at baseline either 3 or 7 days after baseline. Results showed no correlation between 8-OHdG/deoxyguanosine ratios leukocytes any point regardless harvest time. had much higher but displayed 25.6% reduction level (P < 0.04) Leukocytes heparin similar 8OHdG/deoxyguanosine ratios; however, was preferred, it produced clean nuclear pellet without hemoglobin contamination, results less variable. This assay shows within subject stability over both increasing probability that small intervention differences can detected subjects. Buccal provide an accessible pool epithelial represents levels than leukocytes.
