Monitoring and rapid quantification of total carotenoids in Rhodotorula glutinis cells using laser tweezers Raman spectroscopy

摘要: Rhodotorula glutinis is known to accumulate large amounts of carotenoids under certain culture conditions, which have very important industrial applications. So far, the molecular mechanism regulating carotenogenesis still not well understood. To better understand process, it requires methods that can detect rapidly and reliably in single live cells. In this paper, a method based on laser tweezers Raman spectroscopy (LTRS) was developed directly carotenoids, as other biological molecules R. The data showed accumulation lipids occurred mainly late exponential stationary phases when cell growth inhibited by nutrient limitation. Meanwhile, carotenoid concentration changed together with nucleic acids, increased first phase decreased last culture. These demonstrate LTRS rapid, convenient, reliable study process vivo.
