摘要: Recently, we reported that the loss of a telomeric region chromosome 8 in Chinese Hamster Ovary (CHO) cells correlates with higher recombinant productivities. New cell lines lacking this region, called CHO-C8DEL, showed several advantages during line generation and for production proteins (Ritter et al., 2016, Biotechnol Bioeng). Here, performed knock-down knock-out experiments genes located within to identify causing observed phenotypes CHO-C8DEL lines. We present evidence or reduced expression gene C12orf35 is responsible productivities shorter recovery times selection pressure. These effects are mediated by increased levels mRNA exogenes heavy chain (HC) light (LC) as well dihydrofolate reductase (DHFR) neomycin phosphotransferase (Neo) stable antibodies. Biotechnol.
