摘要: C-reactive protein (CRP) is an acute phase serum in man which binds to phosphocholine (PC) a calcium-dependent manner. CRP has been shown bind chromatin and nucleosome core particles. However, does not DNA there conflicting evidence regarding the binding of histones. In present study, was confirmed by ELISA using bound microtiter wells. When depleted histone H1 used same assay, no detected. Similar results were observed competitive inhibition ELISA. These indicate important role for chromatin. Further studies done characterize purified individual histones demonstrated first blotting. Calf thymus separated on 15% SDS-polyacrylamide gel, transferred nitrocellulose, probed with 125I-CRP. H2A lesser extent H2B. Non-specific H3 seen H4 observed. tested Essentially identical those obtained H2A-H2B complex as well reactivity trypsin-resistant fragments H2A, H2B, H3. By blotting all reactions blocked PC EDTA indicating through PC-binding site CRP. further nature show that presence required binding.
