Design and Characterization of a DNA Vaccine Based on Spike with Consensus Nucleotide Sequence against Infectious Bronchitis Virus

摘要: Avian coronavirus infectious bronchitis virus (IBV) causes severe economic losses in the poultry industry, but its control is hampered by continuous emergence of new genotypes and lack cross-protection among different IBV genotypes. We designed a immunogen based on spike with consensus nucleotide sequence (S_con) that may overcome extraordinary genetic diversity IBV. S_con was cloned into pVAX1 vector to form DNA vaccine, pV-S_con. pV-S_con could be correctly expressed HD11 cells corresponding post-translational modification, induced neutralizing antibody response Vero-cell-adapted strain Beaudette (p65) mice. To further evaluate immunogenicity, specific-pathogen-free (SPF) chickens were immunized plasmid compared H120 vaccine. Detection IBV-specific antibodies cell cytokines (IL-4 IFN-γ) indicated vaccination efficiently both humoral cellular immune responses. After challenge heterologous M41, shedding loading tissues significantly reduced homologous vaccine H120. Thus, promising candidate for IBV, approach an appealing method design viruses high variability.
