The porphyrin–fullerene nanoparticles to promote the ATP overproduction in myocardium: 25Mg2+-magnetic isotope effect

摘要: Abstract This is a first case ever reported on the fullerene-based low toxic nanocationite particles (porphyrin adducts of cyclohexyl fullerene-C60) designed for targeted delivery paramagnetic magnesium stable isotope to heart muscle providing sharp clinical effect close about 80% recovery tissue hypoxia symptoms in less than 24 h after single injection (0.03–0.1 LD50). A whole principle this therapy novel: 25Mg2+-magnetic selectively stimulates ATP overproduction oxygen-depleted cells due 25Mg2+ released by nanoparticles. Being membranotropic cationites, these “smart nanoparticles” release overactivating cations only response metabolic acidic shift. The resulting positive changes cell energy metabolism may help prevent and/or treat local myocardial hypoxic disorders and, hence, protect from serious damage vast variety hypoxia-caused situations including both doxorubicin and 1-methylnicotineamide cardiotoxic side effects. Both pharmacokinetics pharmacodynamics drug proposed make it suitable safe efficient administration either or multi-injection (acute chronic) therapeutic schemes.
